For translation or more information on this topic, please contact author.<\/h4>\n<h2>Souhrn<\/h2>\n<p>Tento p\u0159\u00edsp\u011bvek p\u0159edkl\u00e1d\u00e1 praktick\u00e9 zku\u0161enosti s\u00a0chromogenn\u00edm m\u00e9diem CCA, kter\u00e9 se podle novely normy \u010cSN EN ISO\u00a09308-1 pou\u017e\u00edv\u00e1 ke stanoven\u00ed koliformn\u00edch bakteri\u00ed a\u00a0<em>E. coli<\/em> ve vod\u00e1ch s\u00a0n\u00edzk\u00fdm obsahem doprovodn\u00e9 mikrofl\u00f3ry. Jedn\u00e1 se o\u00a0vysoce citlivou metodu, vhodnou pouze pro vody s\u00a0n\u00edzk\u00fdm obsahem doprovodn\u00e9 mikrofl\u00f3ry, p\u0159edev\u0161\u00edm tedy pro dezinfikovan\u00e9 vody (upraven\u00e1 pitn\u00e1 voda, baz\u00e9ny). U\u00a0ostatn\u00edch typ\u016f vod (m\u011blk\u00e9 soukrom\u00e9 studny, povrchov\u00e9 vody, zejm\u00e9na stojat\u00e9\u00a0apod.) je jej\u00ed vyu\u017eit\u00ed sporn\u00e9, p\u0159edev\u0161\u00edm pro vysok\u00fd n\u00e1r\u016fst doprovodn\u00e9 mikrofl\u00f3ry. Tato metoda byla v\u00a0minul\u00fdch letech testov\u00e1na pro p\u0159\u00edpadn\u00e9 pou\u017eit\u00ed v\u00a0p\u0159\u00edrodn\u00edch koupac\u00edch vod\u00e1ch a\u00a0vzhledem k\u00a0n\u00edzk\u00e9 selektivit\u011b (viz\u00a0d\u00e1le) byla vyhodnocena pro tento typ vod jako nevyhovuj\u00edc\u00ed [2].<\/p>\n<p>Koliformn\u00ed bakterie jsou podle t\u00e9to normy skupina gramnegativn\u00edch ty\u010dinek netvo\u0159\u00edc\u00edch spory s\u00a0negativn\u00edm cytochromoxid\u00e1zov\u00fdm testem, kter\u00e9 tvo\u0159\u00ed za aerobn\u00edch podm\u00ednek kolonie b\u011bhem 24\u00a0hodin na\u00a0selektivn\u00edm diferencia\u010dn\u00edm m\u00e9diu p\u0159i\u00a0kultiva\u010dn\u00ed teplot\u011b (36 \u00b1 2) \u00b0C. Koliformn\u00ed bakterie vykazuj\u00ed aktivitu \u03b2-D-galaktosid\u00e1zy a\u00a0<em>E. coli<\/em> je ta koliformn\u00ed bakterie, kter\u00e1 z\u00e1rove\u0148 vykazuje aktivitu \u03b2-D-glukuronid\u00e1zy. V\u00fdznamn\u00e1 je zm\u011bna \u201edetek\u010dn\u00edho syst\u00e9mu\u201c koliformn\u00edch bakteri\u00ed a\u00a0<em>E. col<\/em>i oproti d\u0159\u00edv\u011bj\u0161\u00ed metod\u011b zalo\u017een\u00e9 na fermentaci lakt\u00f3zy (koliformn\u00ed bakterie) a\u00a0tvorb\u011b indolu (<em>E. col<\/em>i). Aktivitu \u03b2-D-galaktosid\u00e1zy vykazuje mnohem \u0161ir\u0161\u00ed spektrum bakteri\u00ed (tento enzym \u0161t\u011bp\u00ed lakt\u00f3zu na galakt\u00f3zu a\u00a0gluk\u00f3zu, kter\u00e1 d\u00e1le m\u016f\u017ee a\u00a0nemus\u00ed b\u00fdt fermentov\u00e1na na kysel\u00e9 produkty); podrobn\u011b jsou biochemick\u00e9 vlastnosti jednotliv\u00fdch druh\u016f bakteri\u00ed z\u00a0\u010deledi <em>Enterobacteriaceae<\/em> uvedeny v\u00a0Bergey\u2019s\u00a0Manual of Determinative Microbiology [3]. Po\u010det koliformn\u00edch bakteri\u00ed detekovan\u00fdch na chromogenn\u00edm m\u00e9diu tak b\u00fdv\u00e1 v\u00fdznamn\u011b vy\u0161\u0161\u00ed, ne\u017e byl jejich z\u00e1chyt na d\u0159\u00edve pou\u017e\u00edvan\u00e9m lakt\u00f3zov\u00e9m TTC agaru s\u00a0tergitolem, a\u00a0v\u00fdsledky proto nekoreluj\u00ed. Total coliforms, <em>E. coli<\/em> and background microflora grown on CCA<\/h6>\n<h2>Metodika<\/h2>\n<p>Koliformn\u00ed bakterie a<em>\u00a0E. coli<\/em> se stanovuj\u00ed membr\u00e1novou filtrac\u00ed (v\u011bt\u0161inou se filtruje objem 100\u2008ml vzorku), membr\u00e1nov\u00e9 filtry maj\u00ed porozitu 0,45\u2008\u03bcm. Filtr se um\u00edst\u00ed na chromogenn\u00ed m\u00e9dium (Chromogenic-Coliform Agar) a\u00a0kultivuje se (21 \u00b1 3) hodiny p\u0159i (36 \u00b1 2) \u00b0C. Slo\u017een\u00ed Chromogenic-Coliform Agaru (d\u00e1le CCA) je uvedeno v\u00a0<em>tabulce 1<\/em>. CCA je b\u011b\u017en\u011b komer\u010dn\u011b dostupn\u00fd. P\u0159\u00edprava CCA je velmi jednoduch\u00e1, nav\u00e1\u017e\u00ed se p\u0159\u00edslu\u0161n\u00e9 mno\u017estv\u00ed pr\u00e1\u0161ku, resp. granul\u00ed, a\u00a0rozpust\u00ed se v\u00a0destilovan\u00e9 vod\u011b. Nep\u0159id\u00e1vaj\u00ed se \u017e\u00e1dn\u00e9 dal\u0161\u00ed p\u0159\u00eddavky. Nesm\u00ed se autokl\u00e1vovat. Po zchlazen\u00ed na 45\u201350\u2008\u00b0C se agarov\u00e9 m\u00e9dium steriln\u011b rozleje do Petriho misek. PH m\u00e1 b\u00fdt 6,8 \u00b1 0,2 p\u0159i 25\u2008\u00b0C (podle na\u0161ich zku\u0161enost\u00ed deklarovan\u00e9 pH v\u017edy sed\u011blo, p\u0159esto je nutn\u00e9 u\u00a0ka\u017ed\u00e9 \u0161ar\u017ee prov\u00e9st kontrolu). M\u00e9dium je velmi st\u00e1l\u00e9, uva\u0159en\u00e9 vydr\u017e\u00ed minim\u00e1ln\u011b m\u011bs\u00edc, jen je nutn\u00e1 ochrana p\u0159ed jeho vyschnut\u00edm. Doprovodn\u00e1 mikrofl\u00f3ra roste ve form\u011b bezbarv\u00fdch koloni\u00ed. Koliformn\u00ed bakterie je nutn\u00e9 potvrdit pomoc\u00ed oxid\u00e1zov\u00e9ho testu (koliformn\u00ed bakterie jsou oxid\u00e1za negativn\u00ed), kter\u00fd norma p\u0159ipou\u0161t\u00ed prov\u00e1d\u011bt p\u0159\u00edmo na membr\u00e1nov\u00e9m filtru. P\u0159i p\u0159\u00edpadn\u00e9m p\u0159eo\u010dkov\u00e1n\u00ed koloni\u00ed na neselektivn\u00ed agar (pro dal\u0161\u00ed konfirmaci) je nutn\u00e9 kolonie pe\u010dliv\u011b p\u0159e\u010distit, aby se v\u017edy pracovalo s\u00a0\u010distou kulturou. Pro p\u0159\u00edpadnou dal\u0161\u00ed konfirmaci, resp. pro potvrzen\u00ed aktivity enzym\u016f \u03b2-D-galaktosid\u00e1zy a\u00a0\u03b2-D-glukuronid\u00e1zy, lze vyu\u017e\u00edt nap\u0159.\u00a0ONP test a\u00a0GLR test (Erba, Lachema). Tyto testy lze vyu\u017e\u00edt i\u00a0pro stanoven\u00ed citlivosti a\u00a0specifi\u010dnosti p\u0159\u00edslu\u0161n\u00fdch reakc\u00ed na CCA. P\u0159i v\u00fdpo\u010dtu v\u00fdsledk\u016f je nutn\u00e9 m\u00edt na pam\u011bti d\u016fle\u017eit\u00fd fakt a\u00a0to, \u017ee i\u00a0<em>E. coli<\/em> pat\u0159\u00ed mezi koliformn\u00ed bakterie, a\u00a0mus\u00ed se tud\u00ed\u017e do jejich po\u010dtu p\u0159i\u010d\u00edst:<\/p>\n<ul>\n<li>po\u010det<em> E. coli<\/em> = po\u010det modr\u00fdch a\u017e modrofialov\u00fdch koloni\u00ed,<\/li>\n<li>po\u010det koliformn\u00edch bakteri\u00ed = po\u010det r\u016f\u017eov\u00fdch a\u017e \u010derven\u00fdch koloni\u00ed s\u00a0negativn\u00edm oxid\u00e1zov\u00fdm testem + po\u010det modr\u00fdch a\u017e modrofialov\u00fdch koloni\u00ed.<\/li>\n<\/ul>\n<h2>V\u00fdsledky a\u00a0diskuse<\/h2>\n<p>Byly testov\u00e1ny des\u00edtky m\u00edrn\u011b organicky zne\u010di\u0161t\u011bn\u00fdch vod, p\u0159edev\u0161\u00edm za \u00fa\u010delem z\u00edsk\u00e1n\u00ed charakteristik metody stanoven\u00ed koliformn\u00edch bakteri\u00ed a\u00a0<em>E. coli<\/em> na CCA. P\u0159esto\u017ee byly vzorky paraleln\u011b testov\u00e1ny na p\u016fvodn\u00edm lakt\u00f3zov\u00e9m TTC agaru s\u00a0tergitolem (\u010cSN EN ISO\u00a09308-1, 2001), v\u00fdsledky nevykazovaly \u017e\u00e1dnou korelaci vzhledem k\u00a0odli\u0161n\u00e9mu principu obou m\u00e9di\u00ed. V\u00a0<em>tabulce 2<\/em> jsou uvedeny v\u00fdsledky Zkou\u0161en\u00ed zp\u016fsobilosti (ZZ) organizovan\u00e9ho ASLAB\u00a0\u2013 OR-MB-1\/15 (vzorky 1\u20135 jsou vzorky, kter\u00e9 slou\u017eily k\u00a0ov\u011b\u0159en\u00ed homogenity v\u00a0laborato\u0159i V\u00daV TGM,\u00a0v.\u2009v.\u2009i.), vzta\u017en\u00e1 hodnota pot\u00e9 byla ur\u010dena ze souboru v\u0161ech \u00fa\u010dastn\u00edk\u016f (KTJ\/ml, n = 19).<\/p>\n<p>Z\u00a0dal\u0161\u00edch charakteristik byly stanoveny produktivita (viz\u00a0<em>obr.\u00a02<\/em>), reprodukovatelnost v\u00a0po\u010d\u00edt\u00e1n\u00ed a\u00a0pod\u00edl fale\u0161n\u011b pozitivn\u00edch a\u00a0fale\u0161n\u011b negativn\u00edch v\u00fdsledk\u016f. Produktivita byla stanovena pomoc\u00ed kmen\u016f <em>E. coli<\/em> WDCM 00013 (CCM 3954) a\u00a0<em>Serratia rubidae<\/em> (CCM 4684), kter\u00e1 nen\u00ed uvedena v\u00a0norm\u011b \u010cSN EN ISO\u00a011133. Dal\u0161\u00ed kmeny, kter\u00e9 jsou p\u0159edeps\u00e1ny ve v\u00fd\u0161e uveden\u00e9 norm\u011b (<em>Enterobacter aerogenes<\/em> WDCM 00175, nebo <em>Citrobacter freundii<\/em> WDCM 00006), nebyly v\u00a0dob\u011b testov\u00e1n\u00ed je\u0161t\u011b k\u00a0dispozici.<\/p>\n<p>V\u00a0informativn\u00ed p\u0159\u00edloze C normy \u010cSN EN ISO\u00a09308-1 [1] je uvedena p\u0159edepsan\u00e1 minim\u00e1ln\u00ed produktivita (v\u00fdt\u011b\u017enost) &gt; 70\u00a0%, a\u00a0na\u0161e testov\u00e1n\u00ed toto hodnotu v\u00fdznamn\u011b p\u0159ekro\u010dilo. Produktivita pro kmen <em>E. coli<\/em> (CCM 3954) byla na CCA od firmy Merck 100\u00a0%, na CCA od firmy Himedia 87,41\u00a0%; produktivita pro kmen <em>Serratia rubidae<\/em>, jako\u017eto z\u00e1stupce koliformn\u00edch bakteri\u00ed (CCM 4684), byla na CCA od firmy Merck 83,82\u00a0% a\u00a0na CCA od firmy Himedia 100\u00a0%.<\/p>\n<h5>Tabulka 2. Aktivita \u03b2-D-glukuronid\u00e1zy byla ov\u011b\u0159ena pomoc\u00ed fluorogenn\u00edho substr\u00e1tu (4-methylumbelliferyl-\u03b2\u2011D-glukuronid), a\u00a0v\u00a098\u00a0% se v\u00fdsledky shodovaly (n = 50). Aktivita \u03b2-D-galaktosid\u00e1zy byla konfirmov\u00e1na ONP testem. V\u00ednov\u00e9 kolonie byly 100% konfirmov\u00e1ny jako \u03b2-D-galaktosid\u00e1za pozitivn\u00ed (0\u00a0% fale\u0161n\u011b pozitivn\u00edch v\u00fdsledk\u016f), u\u00a0sv\u011btl\u00fdch koloni\u00ed byly zaznamen\u00e1ny fale\u0161n\u011b negativn\u00ed v\u00fdsledky v\u00a09\u00a0% (91\u00a0% sv\u011btl\u00fdch koloni\u00ed bylo konfirmov\u00e1no jako\u00a0\u03b2-D-galaktosid\u00e1za negativn\u00ed). D\u00e1le byly ov\u011b\u0159ov\u00e1ny oxid\u00e1za negativn\u00ed kolonie testovan\u00e9 p\u0159\u00edmo na membr\u00e1nov\u00e9m filtru a\u00a0po jejich p\u0159e\u010di\u0161t\u011bn\u00ed byla negativn\u00ed reakce potvrzena v\u00a090\u00a0%.<\/p>\n<p>Mezi hlavn\u00ed v\u00fdhody stanoven\u00ed koliformn\u00edch bakteri\u00ed na CCA agaru pat\u0159\u00ed skute\u010dnost, \u017ee v\u00fdsledky stanoven\u00ed jak koliformn\u00edch bakteri\u00ed, tak <em>E. coli<\/em> jsou k\u00a0dispozici ji\u017e 24\u00a0hodin po nao\u010dkov\u00e1n\u00ed a\u00a0nen\u00ed t\u0159eba \u017e\u00e1dn\u00e9 dal\u0161\u00ed p\u0159eo\u010dkov\u00e1n\u00ed kmen\u016f na neselektivn\u00ed agar. Z\u00e1rove\u0148 norma p\u0159ipou\u0161t\u00ed proveden\u00ed oxid\u00e1zov\u00e9ho testu p\u0159\u00edmo po p\u0159enesen\u00ed membr\u00e1nov\u00e9ho filtru na podlo\u017eku nasycenou oxid\u00e1zov\u00fdm \u010dinidlem. Krom\u011b tak pracn\u00e9ho p\u0159eo\u010dkov\u00e1n\u00ed a\u00a0p\u0159e\u010di\u0161\u0165ov\u00e1n\u00ed koloni\u00ed odpadne i\u00a0n\u00e1sledn\u00fd p\u0159epo\u010det na p\u016fvodn\u00ed po\u010det presumptivn\u00edch koloni\u00ed, \u010d\u00edm\u017e se stanoven\u00ed nejenom zrychl\u00ed, ale i\u00a0zp\u0159esn\u00ed.<\/p>\n<p>Nev\u00fdhodou stanoven\u00ed je velk\u00fd z\u00e1chyt doprovodn\u00e9 mikrofl\u00f3ry, co\u017e znemo\u017e\u0148uje vyu\u017eit\u00ed k\u00a0jin\u00e9mu ne\u017e deklarovan\u00e9mu \u00fa\u010delu (vody s\u00a0n\u00edzk\u00fdm obsahem doprovodn\u00e9 mikrofl\u00f3ry). Dvacet druh\u016f z\u00a0\u010deledi <em>Enterobacteriaceae<\/em> (zahrnuj\u00edc\u00ed rody <em>Cedecea<\/em>, <em>Enterobacter<\/em>, <em>Erwinia<\/em>, <em>Escherichia<\/em>, <em>Hafnia<\/em>, <em>Salmonella<\/em> (<em>S. arizona<\/em>), <em>Shigella<\/em> (<em>S. sonnei<\/em>), <em>Serratia<\/em>, <em>Pantoea<\/em>, <em>Yersinia<\/em>), kter\u00e9 i\u00a0kdy\u017e jsou v\u00a090\u2013100\u00a0% kmen\u016f \u03b2-D-galaktosid\u00e1za pozitivn\u00ed, lakt\u00f3zu nefermentuj\u00ed (dal\u0161\u00ed druhy jsou \u03b2-D-galaktosid\u00e1za pozitivn\u00ed variabiln\u00ed a\u00a0lakt\u00f3za negativn\u00ed). V\u00fd\u0161e uveden\u00e9 skute\u010dnosti skupinu koliformn\u00edch bakteri\u00ed posunuj\u00ed v\u00fdznamn\u011b v\u00edce od fek\u00e1ln\u00edho k\u00a0obecn\u00e9mu zne\u010di\u0161t\u011bn\u00ed a\u00a0mal\u00e9 nedezinfikovan\u00e9 studny prakticky nemaj\u00ed \u0161anci limit pro koliformn\u00ed bakterie 0 KTJ\/100\u2008ml splnit.<\/p>\n<p>Dal\u0161\u00ed malou, ale opticky v\u00fdraznou skupinou jsou kolonie, kter\u00e9 rostou jako tyrkysov\u00e9, v\u00a0n\u011bkter\u00fdch p\u0159\u00edpadech velmi drobn\u00e9 (kter\u00e9 v\u011bt\u0161inou nevyrostou po p\u0159eo\u010dkov\u00e1n\u00ed na neselektivn\u00ed agar), v\u00a0dal\u0161\u00edch p\u0159\u00edpadech rostou jako kolonie b\u011b\u017en\u00e9 velikosti (v\u00a0pr\u016fm\u011bru 1\u2008mm). Podle firemn\u00edch materi\u00e1l\u016f zve\u0159ejn\u011bn\u00fdch na internetu (Merck, VWR Chemicals\u00a0apod.) se jedn\u00e1 o\u00a0dal\u0161\u00ed gramnegativn\u00ed bakterie (doprovodn\u00e1 mikrofl\u00f3ra), \u03b2-D-glukuronid\u00e1za pozitivn\u00ed. Podle na\u0161ich praktick\u00fdch zku\u0161enost\u00ed \u0161lo bu\u010f o\u00a0oxid\u00e1za pozitivn\u00ed kmeny, nebo o\u00a0\u03b2-D-galaktosid\u00e1za negativn\u00ed <em>E. coli<\/em>, p\u0159\u00edpadn\u011b o\u00a0intermedi\u00e1ln\u00ed kmeny z\u00a0oblasti okolo rod\u016f <em>Shigella<\/em> a\u00a0<em>Yersinia<\/em>. V\u00a0n\u011bkter\u00fdch p\u0159\u00edpadech byly konfirmov\u00e1ny i\u00a0\u03b2-D-galaktosid\u00e1za pozitivn\u00ed kmeny.<\/p>\n<h2>Z\u00e1v\u011br<\/h2>\n<p>V\u00fdhody (pop\u0159.\u00a0nev\u00fdhody) vyu\u017eit\u00ed CCA v\u00a0mikrobiologick\u00e9\u00a0praxi uk\u00e1\u017ee \u010das, nicm\u00e9n\u011b u\u017e te\u010f se ukazuje, \u017ee jeho vyu\u017eit\u00ed je vhodn\u00e9 pouze pro vody s\u00a0n\u00edzk\u00fdm obsahem doprovodn\u00e9 mikrofl\u00f3ry (p\u0159edev\u0161\u00edm pro dezinfikovan\u00e9 pitn\u00e9, pop\u0159.\u00a0baz\u00e9nov\u00e9 vody). V\u00a0tomto p\u0159\u00edpad\u011b se jedn\u00e1 o\u00a0stanoven\u00ed rychl\u00e9 (b\u011bhem 24\u00a0hodin jsou k\u00a0dispozici kompletn\u00ed v\u00fdsledky) a\u00a0velmi citliv\u00e9.
